This solution is an interpretation of an experiment that measures hapten-specific B cell response by limiting dilution assay. The explanation begins with a brief background on antigen-antibody response and the limiting dilution method to evaluate it. The effect of pre-exposure to hapten and the generation of hapten-specific B cells is detailed. The benefit of this pre-exposure on antibody response during re-exposure to hapten is the crux of the experiment in this solution. We also discuss the effects of interleukins under these experimental conditions.

The table (which is what I assume you refer to as graph) shows the effect of memory B cell precursors. As background to explaining this assay, I will spend a few lines explaining the antibody response to the hapten TNP, for example. When the body is exposed to TNP, antigen-presenting cells (APCs) present the hapten in a recognizable form to B cells. In the presence of helper T cells, the B cells become plasma cells and secrete antibody directed against the antigen.

In the limiting dilution method, B cells are harvested from the spleens of mice and fewer and fewer B cells are cultured in the presence of the hapten to determine what minimum number (or the limiting dilution) of B cells are required to generate antibody response against the hapten. The magnitude of the response is crudely determined by measuring the number of ...
