The US Food and Drug administration uses safety and efficacy as the two most important criteria to evaluate any drug. A drug can be highly effective but also unsafe. A drug response curve is a technique to evaluate the relationship between its dose and its desired clinical response or toxicity so that its safe and effective use can be determined [1]. Generally, the biological response increases along with increasing concentration of the drug until it reaches a plateau [2]. Examples of a response include enzyme activity, change in heart rate or contraction of a muscle [3]. This plot of the dose-response curve includes concentration of the drug on the x-axis and response on the y-axis. The shape of this curve can be described as rectangular-hyperbola [4]. It is more common to plot the log of concentrations to include a large range for the dose. On this semi-logarithmic plot, the curve becomes sigmoidal [4]. Uses of a dose-response curve include estimating the starting dose, establishing the maximum dose beyond which there is no additional benefit and establishing the therapeutic range.
