Experiment to determine the affinity for competitive antagonists at cholinoceptors in isolated tissues

All the information is in the attached files.

The first 3 files provide a description of what the experiment was about, how we carried it out, and theory etc. I have not been able to attach these files however, can email them on request.

The calculations file are the calculations for the experiment and you need to use these to justofy results etc.

What i want you to do is look at the attachment "write-up guideline" and provide a detailed solution and response to the RESULTS, DISCUSSION & A VERY BRIEF ADDITONAL SECTION areas.

For the calculation of your data, I converted the concentration of catechol to nM. This makes the x-axis of your graphs more reasonable looking.

If you look at the attached Excel file, you will see all of the data displayed in nice columnar format. In addition, in the "Plots" worksheet, you will see the plots which combines all of the four data sets into one graph. It should be self-explanatory. The first graph shows absolute tension. This is important for the reason given in the next paragraph. The second graph shows the normalized percentage data. This is useful for the reasons explained in the paragraph below the next.

Notice the following observations which you should be able to write up in your discussion. The maximum tension is independent of whether atropine is present or not. In addition, the maximum tension is independent of the atropine concentration when present. The maximum tension varies anywhere from about 3.8 g to about 4.8 g with no apparent trend ...

Experiment: Affinity for competitive antagonists

Experiment to determine the affinity for competitive antagonists at cholinoceptors in isolated tissues.

All the information is in the attached files. The first 3 files provide a description of what the experiment was about, how we carried it out, and theory etc.

What i want you to do is look at the attachments and the following:

INTRODUCTION: A brief indication of the aims of the experiment, a definition of competitive antagonism, and of the parameters investigated and an indication as to why it is important to be able to determine pA2, kA etc. Include an indication of which subtype of muscuranic receptor is present in the tissue and how it brings about its response when activated.

METHOD: A mention of the general methods (very brief) and anything specific which is not covered in the schedule (eg weight and sex of animals used)
